Abstract : This study included two phases: 1- Epidemiological and survey study 2- Histopathological study. The first phase was about data collection for the percentage of amoebic dysentery disease in Taif by the records of laboratories of 15 health facilities divided into five sectors of the Taif: (North, East, East, west and south). Around 26,244 patients were examined for stool samples. Statistical analysis of these ratios using (SPSS) was performed, It was found that the overall ratio of the disease Amebic dysentery in the province of Taif in 2009 = 1.24%, and for the sectors, the percentage of cases show that the southern sector is the highest. Summer season is higher percentage than other seasons. The month of July in the summer is the highest percentage and the lowest percentage rate in the month of January in the winter. Also a percentage of the disease in males is higher than in females, and for the age groups the ages from 5 to 14 years is the highest percentage. amoebic dysentery surveillance was contributed factors in the spread of amoebic dysentery in Taif province by water, personal hygiene and sanitation . Histological preparations of normal uninfected Mice large intestine viewed by light microscope revealed typical mammalian organization close to that of human. The most commonly infected organ with Entamoeba Histolytica was the Caecum & Rectum from large intestine in Mice. The present study recorded the most obvious pathological changes in the mucous layer of the large intestine. The infection significantly increased the permeability of cell membranes. Vertical gaps within the epithelial lining of the intestines. There were defects in the centrally positioned nuclei. Cell membranes between cells were almost ruptured. This was resulting in laceration and atrophy of cells (Necrosis). Due to formation of amoebic ulcer, the brush borders of Goblet cells were damaged. Signs of amoebic dysentery as revealed by Scanning Electron Microscope were seen as massive numbers of trophozoites seen to cover the mucosa layer resulting in a cobblestone like appearance. All these changes demonstrate the severity of the damage of the tissue as a result of the inflection by Entamoeba Histolytica that may lead to death
