Abstract : Leukemia is cancer of the blood, acute lymphocytic leukemia (ALL) is the type of leukemia that most commonly affects children, L-asparaginase (ASNase) is an important chemotherapeutic agent used for the treatment of (ALL). The purpose of this study was to study the protective role of octreotide drug against the toxic effect of L-ASNase (Elspar) drug on pancreas of albino rats. Seventy adult male albino rats weighing 200 to 240 g were randomly divided into the following groups: Control Group I received normal saline intraperitonealy for 5 days, Group II received a single intraperitoneal injection of ASNase at a dose of 200 IU/kg body weight for 5 days, Group III received octreotide (10µg/Kg/day) subcutaneously 8 hours before the 200 IU of ASNase for 5 days , Group IV received intraperitoneal injection of ASNase at a dose of500 IU/kg body weight for 5 days, Group V received octreotide (10µg/Kg/day) subcutaneously 8 hours before the 500 IU of ASNase for 5 days, Group VI received intraperitoneal injection of ASNase at dose of1000 IU/kg body weight for 5 days, Group VII received octreotide (10µg/Kg/day) subcutaneously 8 hours before the 1000 IU of ASNase for 5 days. Body weights, pancreatic weights, biochemical, histopathological changes of the pancreas were recorded .There was an increase in amylase enzyme in group IV compared to group II and group VI, also histological changes in group VI and IV, the pancreatic acinar cells showed increase of apical zymogen granules, vacuolization and further damage in the form of focal areas of necrosis of pancreatic tissue. On the other hand, minimal change with group II, and some degree of interlobular fibrosis was detected with group VI. Administration of octreotide induced an improvement in the pancreatic structure. From the results obtained in this work, it was concluded that octreotide prevented the development of ASNase-induced pancreatic injury.
