In vitro Evaluation of Gemcitabine Loaded on Microemulsion Formula in Lung Cancer Cells

Abstract : Gemcitabine is an anticancer drug used to treat several cancer diseases. However, because of its poor bioavailability and cytotoxicity, it has several dose limitation aspects. In this study, three microemulsion formulations, containing the surfactant mixture of polyoxyethylene sorbitan monooleate (tween 80) and sorbitan monolaurate (span 20), isopropyl myristate (IPM) as oil phase and and the aqueous phase of distilled water which includes 40% ethanol, were produced. The antitumor activity of the microemulsion formulas were detected in the A549 non-small cell lung cancer cells and compared with HFS human foreskin cells and the cancerous cells of MCF-7 breast cancer and HCT116 colon cancer. The cytotoxicity was assessed through using sulphorhodamine B (SRB) assay and hemolysis activity, while the mechanism of cell death was determined by observing the morphological changes of treated cells under light microscope, identifying apoptosis by using ApopNexin FITC apoptosis detection kit, and viewing the ultrastructure of A549 non-small cell lung cancer by using transmission electron microscope (TEM). It has been found that capsulations of gemcitabine in microemulsion formulations have showed decreased cytotoxicity against the erythrocytes, as demonstrated by the hemolysis assay. The antiproliferative SRB assay revealed that loading gemcitabine in microemulsion formulations had increased cytotoxic effect against A549, MCF-7 and HCT 116 cells. Furthermore, It has been showed that all of the microemulsion formulations induced apoptosis in A549, MCF-7and HCT116 cells, as viewed under the light microscope and detected by ApopNexin FITC assay. According to the ultrastructure study using TEM, it has been demonstrated that gemcitabine and microemulsion formulations, either blank or gemcitabin- loaded, induced autophagocytosis and apoptosis in A549 cells. This study proved that the microemulsion formulations improved the therapeutic potential of gemcitabine as an anti-cancer drug.
