Abstract : Introduction: Metabolic syndrome (MetS) is a cluster of metabolic disorders, including visceral obesity, dyslipidemia, hypertension, and hyperglycemia. MetS is associated with an increased risk of developing cardiovascular disease (CVD) and type-2 diabetes (T2D). Excessive adiposity leads to systemic release of various adipokines, among which the recently discovered adipokine Vaspin. MetS management holds great importance in preventing or delaying the onset of T2D and CVD events. Natural products have potential affects in MetS management with minimal side effects. A unique polyphenol with low molecular weight, Oligonol, was extracted from lychee fruit have beneficial effects in weight reduction and dyslipidemia. Therefore, the aim of the present study was to investigate the effects of Oligonol as an adjunct to lifestyle counseling on serum levels of Vaspin and MetS components among obese Saudi individuals. Subjects and Methods: 201 participants were randomized in double-blind clinical trial. The study groups were given either Oligonol or a placebo for three months. The weight, height, waist circumference (WC), neck circumference (NC), hip circumference (HC), blood pressure of each participant were measured, and fasting blood samples were taken at the baseline and endpoint to measure total cholesterol, high-density lipoprotein cholesterol (HDL-C), triglyceride, and Vaspin levels. Low-density lipoprotein cholesterol (LDL-C) levels, fat%, body mass index (BMI), WC:Ht, NC:Ht and abdominal volume index (AVI) were calculated using equations. Results: 67 participants completed the study (37 in Group A; 30 in Group B). Significant differences were found in the mean values for body weight, BMI, WC, NC, HC, fat%, WC:Ht, NC:Ht, systolic blood pressure and AVI following supplementation in both groups, and differences in responses were noted for the different genders. Vaspin levels showed no significant changes after supplementation in either group. Conclusions: The results indicated that intervention with supplementation in addition to adhering to healthy diet can be useful in reducing some component of MetS include weight, WC, and systolic blood pressure. As well as, an important cardiovascular indicator parameter LDL-C. If Oligonol was found in Group B after unblinding, it can be considered a useful supplement for controlling metabolic dysregulation among obese Saudi people. However, the serum levels of Vaspin showed minimal differences after supplementation and require further investigation keywords: metabolic syndrome, dyslipidemia, Vaspin, polyphenol, Oligonol
