Abstract : Background: Hypertension is a chronic disease which is considered as one of the leading causes of many cardiovascular disorders. In spite of the significant developments in the antihypertensive therapy, serious side effects still exist. Drugs from herbal origin are prescribed widely in the treatment of different aliments because of their effectiveness, fewer side effects and relatively low cost. Objectives: In this study, vasodilating activity of Hypericum revolutum (Vahl) was fully investigated. Methods: Thin layer chromatography and column chromatography techniques were used to isolate the active compounds of the plant. Compounds were then identified using 1H NMR, 13C NMR, HSQC, and HMBC. Biological study was carried out using the isolated artery technique. Results: Cumulative addition of Hypericum revolutum (Vahl) total extract (1-10 μg/ml) led to a relaxation of phenylephrine-precontracted aortae. Bio-guided fractionation revealed that the chloroform fraction is responsible for this effect. The observed vasodilation is endothelial nitric oxide (NO) synthase-dependent as it was blocked by endothelial denudation or N-nitro-L-arginine methyl ester hydrochloride (L-NAME), the NO synthase inhibitor. Chemical investigation of chloroform fraction of Hypericum revolutum (Vahl) gave rise to one new phloroglucinol derivative besides other known xanthones. Conclusion: Hypericum revolutum (Vahl) compounds have inherent ability in vasodilatation and consequently in the treatment of hypertension.
